Melanocortin and neuroendocrine signaling
This category covers two receptor families. Melanocortin receptors span five subtypes with distinct distributions — MC1R in melanocytes, MC3R and MC4R in the central nervous system — and synthetic analogs are studied primarily for subtype selectivity and the cAMP response each subtype produces. Neuropeptide receptors such as the oxytocin receptor sit in a separate family and are studied for their role in social-bonding and stress-response pathways.
Why selectivity is the whole question
A molecule that activates several melanocortin subtypes produces a composite readout that cannot be attributed to any one of them. Subtype-selective binding data is therefore the first thing to look for in the literature on any compound in this category.
